This is a story about two guys, Sameer (Abhishek Bachchan) who is a nurse and Kunal (John Abraham), a photographer. They bump into each other while looking for an apartment to rent and coincidentally head towards the same apartment. However, they are both thrown out because the owner wants to rent the apartment to girls only, since her niece, Neha (Priyanka Chopra) stays in that apartment alone. Sameer gets an idea (when a US Soldier mistakes them to be gay couple and tells them about missing his partner) that if Kunal and he pretend to be gay lovers, then the owner will agree to let them stay. Kunal initially refuses, but agrees out of desperation. Their plan works and they move in. Over a period of time Neha, Kunal and Sameer become good friends, hanging out together and having fun.
One day Neha's boss 'M', short for Murli (Boman Irani) announces that he is resigning his job as the editor-in-chief of Verve magazine and is in charge of replacing himself. In order to impress her boss (who is gay) and bag the job, Neha invites him for dinner at her house telling him that she will introduce him to her gay roommates. The evening turns out to be a comical situation as Sameer's mother finds out that her son might be gay and turns up at their doorstep. Next day at Neha's office M announces Abhimanyu Singh (Bobby Deol) as the next editor-in-chief. Neha, heartbroken returns home where Kunal and Sameer try to console her. Kunal, Sameer and Neha then together work out a difficult project she was given by her boss and Neha becomes a hit at her office. Looking at her success Abhimanyu appoints her as the co-editor-in-chief of Verve.
Meanwhile, in Kunal's absence Sameer takes out Neha out and they share an intimate time. Later, at a mall Sameer tries to tell Neha he loves her and that he's not gay through a trial room door, but turns out Kunal was behind that door and heard everything he said. He becomes angry with Sameer and states that he also loves her. As they try to figure out what to do Kunal says that as tomorrow's Neha's birthday they should go out to this new place he knows, but actually sends him to a male strip bar while he and Neha enjoy an intimate dinner and movie at a park. Sameer comes to know of this and furiously hits Kunal and a fight breaks out which Neha ends. The next day, Abhimanyu asks Neha out to an event and asks her to bring her two friends along. In the event while the trio are enjoying, Abhimanyu enters and sweeps Neha away. Then Kunal and Sameer decide to first eliminate him together. They try telling him to do exact opposite of things that Neha likes (like asking him to wax his chest hair off and wear outlandish dress), but their plan doesn't work. They then set their sights on Abhimanyu's 5-year old son Veer (Shrey Bawa). They try to make Veer hate Neha. The next day, Neha comes to know that Abhi is going to propose her. Neha goes to Kunal and Sameer, and asks them what she should do as she is not even thinking about marriage yet. Both of them ask her not marry him and reveal that they aren't gay and are in love with her. Meanwhile Veer tells Abhimanyu of his fears and cries saying that he doesn't want him to marry Neha. As Neha tearfully turns away from Kunal and Sameer, Abhimanyu hugs her and tells her that they can't be together as Veer isn't comfortable with that. Kunal and Sameer see them hugging and presume them to be engaged. Neha comes home and asks the guys to leave the house.
Later, Kunal and Sameer go to the immigration office to get their residence permit and realize that they miss Neha, so they go to get her at an office party that she is attending. They try explaining to her but she refuses to listen and Abhimanyu enters. Kunal asks when they are going to get married and Neha tells him that they broke up because Veer wasn't comfortable with her. They both tell them the truth and Abhimanyu and Neha furiously try to leave the place. But the guys stop them and get on to the stage and tell the crowd their story and ask them tell Neha to grant forgiveness. The crowd edges them to do stuff like get on the knees and beg to Neha, when one of them asks them to kiss each other . The guys adamantly refuse to do it when Abhimanyu says that since they are very fond of being gay, it is the only way they can be forgiven. Kunal refuses to do it but Sameer convinces him and tries to kiss him but at the end moment he cant do it. As Kunal sees Neha and Abhimanyu starting to leave he forcibly kisses Sameer and Abhimanyu tells Neha that those guys were really her best friends to do this as even he couldn't have done it, even for her. Neha goes to them and they make up. After two months Neha, Kunal and Sameer are sitting in their apartment balcony and Neha asks them if anytime during their act of being gay, they ever felt anything towards each other. They vehemently deny it and Neha leaves apologizing. Sameer and Kunal look at each other and remember their disastrous kiss, and walk away.

Sanjay Singhania (Aamir Khan), a rich businessman who owns a cellular phone company named Air Voice.Kalpana, an advertisement model goes around telling her advertisement crew that she is the love interest of Sanjay in spite of she never meeting him. Sanjay happens to hear about this through the media and wants to settle scores with her. On his way, he catches a fleeting glimpse of a young woman (who happens to be Kalpana) helping handicapped children to cross a street and falls in love with her. He moves on to confront Kalpana at her advertisement studio. He is pleasantly surprised to realize that she is the same helpful woman he saw a while ago. He chooses to hide the fact from her that he is Sanjay, calling himself as Sachin. Kalpana eventually falls in love with him.
Once, while on a train to Mumbai, Kalpana saves a group of young girls from abuse by a group of goons led by Ghajini (Pradeep Rawat). After a few days, the same goons come to Kalpana's house in order to avenge their embarrassment. Incidentally, Sanjay arrives at the scene and fights off the goons, but during the ensuing fight, Kalpana is murdered and Sanjay suffers head injuries. Sanjay survives the blow, but suffers from a short-term amnesia. At any given time, he does not remember anything that happened more than 15 minutes back, although he frequently has flashbacks of Kalpana's murder. He determines to take revenge on those responsible for Kalpana's death. The rest of the story is about how Sanjay, along with a medical college student Sunita (Jiah Khan), goes about finding Ghajini and the goons.
A medical student Sunita is interested in doing a project on Sanjay in order to study human brain. But her professor does not allow her to do so because this was a legal case. In the mean while Sanjay started killing two of the murderers. He used to keep a camera with him which beeps after 15 mins to keep in his memory that he has a camera with him. He takes pictures of those whom he interacts and make notes on the photograph about his relationship with them. He keeps photographs of his apartment, his manager, his doctor etc. The goons or the villain Ghajini finds about Sanjay's attempts to kill him (Ghajini) and when in an unconscious state covers his tattoos and burns his photographs so that he does not regain his memory. However as Sunita learnt more about the story, she decided to help Sanjay. She visits Sanjay in the hospital and shows him his diaries and other evidences regarding Kalpana and her fateful death. Finally Sanjay Singhania takes his revenge by killing Ghajini in the same manner as Ghajini killed Kalpana. In the process, Sunita and Sanjay are placed in the same position that he and Kalpana were placed in before she died and Sanjay recollects the memory of seeing Kalpana on the floor.

Fashion begins in Chandigarh, where aspiring model Meghna Mathur (Priyanka Chopra) expresses her wish to go to Mumbai and pursue her dream of becoming a supermodel. While her dad (Raj Babbar) outrightly denies her permission, her mom (Kiran Juneja) is supportive. Against her dad's wishes, Meghna leaves her house with dreams in her eyes to strike it big in the modeling world. But it is not a path of roses for her. She experiences some major difficulties in her initial days in Mumbai auditioning several times but getting rejected. But she gets a good helping hand with an old acquaintance, Rohit (Ashwin Mushran),an aspiring gay designer who assists big shot designer Vinay Khosla (Harsh Chhaya). She keeps bumping into another struggling model, Maanav (Arjan Bajwa) while making the rounds of modeling agencies for auditions.
She visits Rahul Arora's fashion show where she comes face-to-face with reality of the major people in the industry including the reigning supermodel Shonali Gujral (Kangana Ranaut). She is mocked by the show choreographer Christine and is advised to visit hot-shot photographer Kartik (Rohit Roy) for her portfolio. To pay Kartik's high fee she attends functions and even shoots for a lingerie advertisement, all the while mentored by another smalltime model Janet (Mugdha Godse). Meghna becomes good friends with Maanav and even starts sharing his apartment after she faces accommodation problems. Meanwhile Meghna continues to strive, modeling for various designers as a side-by, all the while, aspiring to be as successful as supermodel Shonali Gujral (Kangana Ranaut)is. Meghna gets noticed by a top official Anisha Roy (Kitu Gidwani) of a major modeling agency Panache who brings her to her boss, Abhijit Sarins (Arbaaz Khan) notice. Abhijit gets impressed by her ambitious nature and go-getter attitude. Panaches face is Shonali but though very successful, her drug abuse starts becoming a problem for her and subsequently for Panache. Abhijit decides to terminate Shonalis contract and ropes in Meghna to replace Shonali as Panache's new face. Shonali warns Meghna that she will just lose herself in all that jazz, like she did.
Meghna becomes an overnight success and is everywhere right from being the showstopper at the biggest fashion shows in town to fashion magazine covers to print ads to TV commercials. The overnight success spoils Meghna and the budding love relationship between her and Manav also comes to an abrupt end. Meanwhile, Janet, an old time acquaintance of Rahul, grabs an opportunity of being his show-stopper. Rahul gets worried as his mother becomes increasingly doubtful of his sexual orientation and asks Janet to marry him though he tells that his relation with his boyfriend will only be compromised to some extent. Shonali, on other hand, gets a wardrobe malfunction at an event and her career comes to an end. Her drug abuse gets worse and she lands up at a rehab centre. Meanwhile, Meghna gets pregnant with Abhijit's child and reluctantly has an abortion due to the conditions in the contract earlier added by Abhijit himself. She tells about their relation to Abhijit's wife and Abhijit terminates Meghna's contract with Panache.
An angered Meghna resorts to alcohol to relieve herself. She gets into a verbal fight with Janet and on the same night attends a rave party where under the influence of cocaine, she unknowingly ends up in bed with a black male escort. Later when she comes to her senses she feels guilty of her act and leaves Mumbai to return to her parents in Chandigarh.
Broken and depressed Meghna lives in Chandigarh for more than a year but she has lost all taste for life including her self-confidence, but encouraged by her father,again goes back to Mumbai. There she befriends Janet again and models for Rohit's show. However, she freezes on stage and gets negative reviews from the whole industry and media. She visits Maanav to ask forgiveness and gets to know that he is engaged. Shonali is shown on TV as a highly disturbed alcoholic on the streets. Meghna brings her home and takes it as a challenge to rehabilitate Shonali. She also gets an offer from Rahul to be his show-stopper but refuses only to accept it later when Shonali encourages her. One day before the show Shonali goes missing from Meghna's home. Just before Meghna was to walk the ramp, she gets a call from the police that Shonali has been found dead apparently from a drug overdose. Meghna becomes frigid with sadness and is unable to walk the ramp. But she overcomes her grief to walk the ramp. Thereafter, she is shown breaking down to tears onstage in front of the media. She ultimately acknowledges that her ensuing success is not due to her own efforts to revamp herself, but due to Shonali and Shonali alone, for, she was the one who instilled once again her newly found self confidence and also gave her a self recognition in this so called glamorous world of fashion. Destiny has failed to give Shonali a second chance to recoup her own losses but through manifestation of her blunders,has taught Meghna a lesson to face the wicked world once again with anew vigor. Meghna hence gives up her drinking and smoking habits and takes on the ramps once again as an ideal model. Her career takes a new flight again and she achieves a new pinnacle of success. The movie ends with Meghna shown walking the ramp in Paris.
